Running Anan 10E, Thetis 2.8.11, Firmware 10.3, Protocol 2, Windows10 fully updated...
I am unable to make PTT out, pin 1 of accessory connector, activate on anything other than mic PTT from front panel. PTT out will not activate using, for example, Tune, MOX, and most importantly CAT or CAT/PTT from WSJTX. The problem is that all of these inputs successfully key the rig but not PTT out, which I need for amplifier control besides mic input. I am unable to find relevant setup settings or Community topics.
If anyone could help I would be most grateful. Thanks everyone.

If you load Protocol 1 firmware into the ANAN does it work then?

What happens when you have nothing plugged into the front panel mic connector? Does MOX etc. work then?

With nothing in the front panel TUN, MOX, CAT, CAT+PTT still key the 10E but not PTT out. I also noticed today keying CW will key the 10E but not PTT out.

I think I've got it sorted for you: you are using the wrong pin on the accessory connector as PTT OUT. It seems like you are using the PTT IN pin, which is in parallel with the mic PTT signal. You need to use the correct pin for PTT OUT.

Thanks Scott, that was the problem. Looking at the back of the 10E, I was using the bottom left-hand most "pin1" on the left side of the table AND the red pin number assignments in the User Guide. Reading table numbers on the right side indicated "pin 25" which per the red numbering is the bottom right-hand most pin which works fine. MOST confusing but all's well that ends well. I can't thank you enough for your help.
